<p><span style="font-size: 16px;">Un tipico bioreattore monouso è costituito da una camera di coltivazione sterile in materiale polimerico. Dopo il suo riempimento asettico con il terreno di coltura e la successiva inoculazione con l’organismo da moltiplicare (in genere cellule animali o vegetali), il bioreattore inizia la sua attività. Ultimata la raccolta del prodotto (ad esempi cellule, proteine ricombinanti, metaboliti secondari), il bioreattore viene eliminato. In raffronto ai bireattori in vetro od acciaio, i bioreattori monouso hanno diversi vantaggi: tempi ridotti di preparazione, eliminati i processi di pulizia e sterilizzazione, ridotta possibilità di contaminazione crociata, grande semplicità e flessibilità. Inoltre sono corredati di documentazione di sterilità. Questi sistemi monouso possono essere classificati in sistemi di coltura su piccola scala, bioreattori a “hollow fiber” e bioreattori in sacchetto.</span></p>
<p><span style="text-decoration: underline; font-size: 16px;">- I sistemi su piccola scala</span></p>
<p><span style="font-size: 16px;">Le tradizionali scatole Petri, i matracci, le “roller bottles” sterili sono da sempre utilizzati nei laboratori grazie alla loro semplicità ed all’impiego di strumentazione e sistemi di controllo elementari (incubatori, shakers, etc.) per assicurare condizioni ambientali appropriate per le cellule. Sono tipicamente sistemi produttivi per la moltiplicazione di cellule animali in sospensione od ancorate.</span></p>
<p><span style="font-size: 16px;">Questi sistemi tendono ad essere sostituiti oggigiorno da sistemi di nuova generazione che consentono di ottenere densità cellulari e prodotti in quantità più elevati (&gt;10<sup>7</sup> cellule / ml).</span></p>
<p><span style="font-size: 16px;">Un esempio è costituito dal sistema “Miniperm” della Greiner-Bio-One. <b><br />
</b></span></p>
<p><span style="font-size: 16px;">Un altro sistema monouso sterile, pronto all’uso è il “Fibrastage” della New Brunswick Scientific. <b><br />
</b></span></p>
<p><span style="text-decoration: underline; font-size: 16px;">- Conclusioni</span></p>
<p><span style="font-size: 16px;">I vantaggi dei bioreattori monouso si possono così riassumere:</span></p>
<p><span style="font-size: 16px;">1. maggiore resa in densità cellulare e prodotti del metabolismo</span></p>
<p><span style="font-size: 16px;">2. ridotta possibilità di contaminazione crociata</span></p>
<p><span style="font-size: 16px;">3. riduzione dei tempi di preparazione</span></p>
<p><span style="font-size: 16px;">4. eliminazione dei processi di pulizia, sterilizzazione</span></p>
<p><span style="font-size: 16px;">5. semplicità e flessibilità.</span></p>

La scoperta di un meccanismo alla base di questo depauperamento apre la strada allo sviluppo di nuove strategie terapeutiche contro l&#8217;indebolimento muscolare, anche quello legato alle varie forme di distrofia <em>(red)</em><b></b></span></p>
<p><span style="font-size: 16px;">L&#8217;indebolimento muscolare dovuto all&#8217;età dipende da una perdita di efficienza delle cellule preposte alla riparazione e alla ricrescita dei tessuti muscolari &#8211; le cosiddette cellule satelliti &#8211; che a sua volta è legata a un progressivo aumento dell&#8217;attivazione di specifiche vie di segnalazione cellulare. E&#8217; questa la conclusione di due ricerche pubblicate su “Nature Medicine”, una condotta all&#8217;Università di Ottawa e l&#8217;altra da ricercatori della Fondazione Santa Lucia di Roma e del Sanford-Burnham Medical Research Institute a La Jolla, in California. </span></p>
<p><span style="font-size: 16px;"> Le cellule satelliti sono una particolare sottoclasse di staminali adulte che, a differenza delle altre staminali, non sono pluripotenti ma unipotenti, ossia capaci di differenziarsi solo nei miociti dei muscoli scheletrici. Nel processo di ricostituzione del tessuto muscolare, queste cellule satelliti devono dividersi in modo asimmetrico, in modo che in una parte delle cellule figlie si inneschi la differenziazione finale in cellule muscolari mature, mentre un numero sufficiente di altre cellule figlie continui a mantenere le caratteristiche rigenerative delle genitrici.</span></p>

				<content:encoded><![CDATA[<p><span style="font-size: 16px;">Security forces worldwide rely on sophisticated equipment, trained personnel, and detection dogs to safeguard airports and other public areas against terrorist attacks. A revolutionary new electronic chip with nano-sized chemical sensors is about to make their job much easier.</span></p>
<p><span style="font-size: 16px;">The groundbreaking nanotechnology-inspired sensor, devised by Prof. Fernando Patolsky of Tel Aviv Univ.&#8217;s School of Chemistry and Center for Nanoscience and Nanotechnology, and developed by the Herzliya company Tracense, picks up the scent of explosives molecules better than a detection dog&#8217;s nose. Research on the sensor was recently published in the journal <i>Nature Communications</i>.</span></p>
<p><span style="font-size: 16px;">Existing explosives sensors are expensive, bulky and require expert interpretation of the findings. In contrast, the new sensor is mobile, inexpensive, and identifies in real time — and with great accuracy — explosives in the air at concentrations as low as a few molecules per 1,000 trillion.</span></p>
<p><span style="font-size: 16px;"><b>A nano-nose to compete with a dog&#8217;s</b></span></p>
<p><span style="font-size: 16px;">&#8220;Using a single tiny chip that consists of hundreds of supersensitive sensors, we can detect ultra low traces of extremely volatile explosives in air samples, and clearly fingerprint and differentiate them from other non-hazardous materials,&#8221; said Prof. Patolsky, a top researcher in the field of nanotechnology. &#8220;In real time, it detects small molecular species in air down to concentrations of parts-per-quadrillion, which is four to five orders of magnitude more sensitive than any existing technological method, and two to three orders of magnitude more sensitive than a dog&#8217;s nose.</span></p>
<p><span style="font-size: 16px;">&#8220;This chip can also detect improvised explosives, such as TATP (triacetone triperoxide), used in suicide bombing attacks in Israel and abroad,&#8221; Patolsky added.</span></p>
<p><span style="font-size: 16px;">The clusters of nano-sized transistors used in the prototype are extremely sensitive to chemicals, which cause changes in the electrical conductance of the sensors upon surface contact. When just a single molecule of an explosive comes into contact with the sensors, it binds with them, triggering a rapid and accurate mathematical analysis of the material.</span></p>
<p><span style="font-size: 16px;">&#8220;Animals are influenced by mood, weather, state of health and working hours, the oversaturation of olfactory system, and much more,&#8221; says Patolsky. &#8220;They also cannot tell us what they smell. Automatic sensing systems are superior candidates to dogs, working at least as well or better than nature. This is not an easy task, but was achieved through the development of novel technologies such as our sensor.&#8221;</span></p>
<p><span style="font-size: 16px;"><b>A technology for a safer world</b></span></p>
<p><span style="font-size: 16px;">The trace detector, still in prototype, identifies several different types of explosives several meters from the source in real time. It has been tested on the explosives TNT, RDX, and HMX, used in commercial blasting and military applications, as well as peroxide-based explosives like TATP and HMTD. The latter are commonly used in homemade bombs and are very difficult to detect using existing technology.</span></p>
<p><span style="font-size: 16px;">&#8220;Our breakthrough has the potential to change the way hazardous materials are detected, and of course should provide populations with more security,&#8221; says Patolsky. &#8220;The faster, more sensitive detection of tiny amounts of explosives in the air will provide for a better and safer world.&#8221;</span></p>
<p><span style="font-size: 16px;">Tracense has invested over $10 M in research and development of the device since 2007, and expects to go to market next year. Prof. Patolsky and his team of researchers are currently performing multiple and extensive field tests of prototype devices of the sensor.</span></p>

				<content:encoded><![CDATA[<p><span style="font-size: 16px;">Il concetto di microorganismo effettivo è stato sviluppato dal <a title="Microbiologia" href="http://it.wikipedia.org/wiki/Microbiologia">microbiologo</a> <a title="Giappone" href="http://it.wikipedia.org/wiki/Giappone">giapponese</a> <a title="Teruo Higa" href="http://it.wikipedia.org/wiki/Teruo_Higa">Teruo Higa</a>, Professore dell Università di Ryukyus in Okinawa Giappone, che analizzando li comportamento di varie combinazioni di microorganismi ha elaborato il &#8220;principio di prevalenza&#8221;, non pienamente comprovato scientificamente. </span></p>
<p><span style="font-size: 16px;">L&#8217;influenza di una combinazione di alcuni ceppi di microrganismi in <a title="Ecosistema" href="http://it.wikipedia.org/wiki/Ecosistema">ecosistemi</a> come terreni o acque. </span></p>
<p><span style="font-size: 16px;">I microrganismi da lui osservati, se posti in maggioranza, influenzano a catena il comportamento di altri microrganismi e stimolerebbero diverse attività biologiche <a title="Antiossidante" href="http://it.wikipedia.org/wiki/Antiossidante">antiossidanti</a>, da lui denominate &#8220;rigenerative&#8221;, che contrastano le attività biologiche <a title="Ossidazione" href="http://it.wikipedia.org/wiki/Ossidazione">ossidative</a>, da lui chiamate &#8220;degenerative&#8221;.</span></p>
<p><span style="font-size: 16px;"> Teruo Higa, affermò che una combinazione di circa 80 microorganismi è capace di decomporre la materia organica in modo da &#8220;promuovere la vita&#8221;. </span></p>
<p><span style="font-size: 16px;">Ci sarebbero tre gruppi di microorganismi: &#8220;microorganismi positivi&#8221; (che hanno effetto sulla rigenerazione della vita), &#8220;microorganismi negativi&#8221; (che decompongono) e &#8220;microorganismo opportunisti&#8221;. </span></p>
<p><span style="font-size: 16px;">In ogni <a title="Bioma" href="http://it.wikipedia.org/wiki/Bioma">bioma</a> il rapporto tra &#8220;positivi&#8221; e &#8220;negativi&#8221; è critico dato che i microorganismi opportunisti possono prendere o perdere il sopravvento in determinate condizioni. </span></p>
<p><span style="font-size: 16px;">Higa pensa che è possibile influenzare positivamente il bioma in oggetto fornendo colonie di &#8220;microorganismi positivi&#8221;</span><b></b></p>

<p><span style="font-size: 16px;">Un team di ingegneri olandesi ha pubblicato un articolo in cui mostra una nuova invenzione: un microscopico sperma robotico che può essere controllato con un debole campo magnetico. Come per lo sperma reale, il MagnetoSperm ruota la propria coda per nuotare verso il loro obbiettivo.</span></p>
<p><span style="font-size: 16px;"> A differenza degli spermatozoi reali sono hanno una testa ed una coda ma sono lunghi circa 6 volte di più.</span></p>

<p><span style="font-size: 16px;">Realizzati in un polimero hanno la testa ricoperta di metallo il che permette agli scienziati di controllare i nanobot in modo piuttosto preciso grazie ad un campo magnetico. E sebbene non siano così veloci quanto gli spermatozoi biologici sono di grande aiuto, dicono gli scienziati, per portare farmaci ad uno specifico target così come essere un nuovo metodo di fertilizzazione in vitro.</span></p>
<p><span style="font-size: 16px;"> Ora l&#8217;obbiettivo renderli più piccoli e più veloci. Magari potrebbero essere persino utili per funzionare come lavoratori microscopici per assemblare altre nanotecnologie.</span></p>

Tuttavia queste tecniche, che in via teorica sarebbero più facili da applicare ai Neanderthal rispetto ad altri organismi estinti, hanno il loro più reale campo d&#8217;applicazione nella prevenzione dell&#8217;estinzione, con l&#8217;arricchimento della diversità genetica di specie in pericolo.</span></p>

<p><span style="font-size: 16px;"> <strong>The finding</strong></span><br />
<span style="font-size: 16px;"> Genetically engineered microorganisms could be used to produce fuels and industrial products, but some microbes’ metabolisms get in the way, degrading or blocking synthesis of the products scientists want. By keeping an extreme heat-loving microorganism below its accustomed temperature, University of Georgia biochemist Michael Adams and colleagues deactivated many of the hyperthermophile’s own metabolic processes and engineered a pathway that could ultimately be manipulated to produce liquid fuel.  </span></p>
<p><span style="font-size: 16px;"> <strong>The methods</strong></span><br />
<span style="font-size: 16px;"> Adams and colleagues took five genes from one archaebacterium (<em>Metallosphaera sedula</em>) that uses hydrogen gas as an energy source to incorporate carbon dioxide into organic compounds, and inserted those genes into the hyperthermophile <em>Pyrococcus furiosus</em>, another archaebacterium. The modified microbe produced 3-hydoxypropionic acid, a valued industrial compound used in acrylic plastics production.</span></p>
<p><span style="font-size: 16px;"> <strong>The innovation</strong></span><br />
<span style="font-size: 16px;"> The hyperthermophile <em>P. furiosus</em> grows best at 100 °C, but the scientists cultured it near 70 °C, a temperature at which “it’s sort of a close-to-inactive bowl of cytoplasm, whereas the genes we engineered into it are maximally active at 70 degrees,” says Adams. By shifting the culturing temperature, the team enabled the engineered microbe to efficiently perform new tasks without interference from its usual metabolic processes.</span></p>
<p><span style="font-size: 16px;"> <strong>The model</strong></span><br />
<span style="font-size: 16px;"> “This is exciting work that extends frontiers in several important ways,” demonstrating both a new way to make useful organic chemicals and a strategy for genetically engineering a heat-loving microbe, says Lee Lynd, a metabolic engineer at Dartmouth College.</span></p>
